Description

One option for treating dementia could be to protect neurons from damage through the use of neuroprotective molecules, such as sAPPα. This thesis used two unbiased receptor identification methods to identify sAPPα and sAPPβ binding partners in neuronal cells with the aim of identifying novel therapeutic targets. Calsyntenin-3 was identified as a novel binding partner which was required for the binding of sAPPα to neuronal cells. Included are four separate files containing the full lists of identified proteins for microbead based and QCM-D based pulldowns using sAPPα and sAPPβ. File #1 contains the proteins identified from pulldowns 1 and 2 using SH-SY5Y cells with sAPPα, sAPPβ or the OKT9 antibody. These samples were analysed through GeLC-MS/MS using an Orbitrap Elite (ThermoFisher Scientific) mass spectrometer. File #2 contains the proteins identified from pulldown 3 using SH-SY5Y cells with sAPPα, sAPPβ or the OKT9 antibody. These samples were analysed through GeLC-MS/MS using a Thermo QE HF with U3000 nanoUPLC mass spectrometer. File #3 contains the proteins identified from human immortalised cell membrane-enriched exract, isolated using QCM-D and sAPPα or sAPPβ. These samples were analysed through GeLC-MS/MS using an Orbitrap Elite (ThermoFisher Scientific) mass spectrometer. File #4 contains the proteins identified from human iPSC-derived neuron cell lysate, isolated using QCM-D and sAPPα, sAPPβ or a negative control. These samples were analysed through GeLC-MS/MS using an Orbitrap Elite (ThermoFisher Scientific) mass spectrometer.
