Throughout my work I have used a multidisciplinary approach to address research questions. These range from molecular biology (e.g. high throughput construct cloning, mutational analysis), cell culture (e.g. bacterial and mammalian systems), recombinant protein purification (e.g. IMAC, SEC), biophysical techniques (e.g. MALS, AUC, SPR, MST, ITC), structural techniques (e.g. SAXS, cryo-EM) and MD modelling (CNS). By bringing together these multiple techniques and corroborating them, it has been possible to answer research questions with much greater accuracy.
Throughout my PhD, SAXs has been my main techniques of interest. I have used several of the BioSAXS beamlines in Europe and have analysed and evaluated data using the main software packages (ATSAS, ScÅtter). I have also developed additional tools for real space rigid body modelling using true MD techniques. More recently cryo-EM has been used at the University of Manchester since the purchasing of a Gatan K2 Summit Direct Detection camera. Data has been analysed using a multitude of EM related software packages (EMAN, Relion).
