    Abstract

Various studies have confirmed that HLA-DRB1*03 is a risk factor for idiopathic inflammatory myositis, but have failed to elucidate any significant genetic differences between PM and DM. Through the UK Myositis Immunogenetic Collaboration, we have collected DNA phenotype data on 244 cases.AIMS: 1. To evaluate the role of HLA-DRB1 & DQB1 genotyping in PM, DM and PM/overlap disease2. To determine the genetic differences of PM and DM in a UK White Caucasian population METHODS: 244 adult UK white patients; PM, n=98 (28 males, 49.7 ± 14.7 years), DM, n=85 (26 males, 45.4 ± 14.7 years), PM/overlap, n=61 (12 males, 47.4 ± 11.5 years), with definite myositis, according to the Tanimoto criteria (J Rheumatol 1995, 22:668-674), were compared with a randomly selected, ethnically matched control group (n=537). DNA was amplified by the polymerase chain reaction (PCR) and HLA-DRB1 genotyping was performed using the Dynal RELITM SSO test.RESULTS: There was an overall difference in the HLA-DRB1 global phenotype frequencies between the whole myositis population and controls (p
