Rapid evaluation of microbial count in river water based on headspace concentration of volatile organic compounds

Abstract

Rapid determination on the microbial count in water will be a useful index to evaluate its safety for daily activities. This study proposes a rapid method to evaluate total microbial count for river water by using the concentration of the volatile organic compounds (VOCs) formed on the headspace as indicator. Water sample was first filtered through a 0.22 micron filter, which liter the filter was incubated in an airtight 2L stainless steel chamber using R2A nutrient broth. The concentration of the VOCs on the headspace over time was found correlating directly to the density of the microbial in the broth, determined using conventional microbial test. The proposed method has showed significant result on the total microbial count using the concentration reading of the VOCs after 6 hr compared to conventional method that required at least a day. This method is not only rapid, but also simple to perform.
